神经生长因子（NGF）是神经营养因子中最早被发现，目前研究最为透彻，具有神经元营养和促突起生长双重生物学功能一种神经细胞生长调节因子，它对中枢及周围神经元发育、分化、生长、再生和功能特性表达均具有重要调控作用。神经生长因子包含α、β、γ三个亚单位，活性区是β亚单位。β-神经生长因子由两个118个氨基酸组成单链通过非共价键结合而成二聚体，与人体神经生长因子结构具有高度同源性，生物效应也无明显种间特异性。NGF与受体结合，通过受体介导内吞机制产生内在化，形成由轴膜包绕、含有NGF、并保持其生物活性小泡，经轴突沿微管逆行转运至胞体，经酪氨酸蛋白激酶、脂酰肌醇钙、内源性环腺苷酸等第二信使体系转导，启动一系列级联反应，对靶细胞某些结构或功能蛋白基因表达进行调控而发挥其生物效应。
人β-神经生长因子，CHO表达重组蛋白是在CHO中国仓鼠卵巢细胞中重组表达的，是由两条糖基化多肽链组成的同源二聚体，每条多肽链包含有120个氨基酸，分子量为16，950Da。该人β-神经生长因子，CHO表达重组蛋白最终由标准的色谱技术纯化出。

溶解：提供的人β-神经生长因子,CHO表达(b NGF CHO)产品为经0.2um孔径过滤灭菌的白色冻干粉末；该冻干粉末是从含人β-神经生长因子,CHO表达(b NGF CHO)的30mM 乙酸钠（pH 5.5），1% HSA溶液中冻干产生的。我们建议加无菌的18MΩ-cm水以配制工作储存液到不低于100ug/ml，后继实验过程中可根据实验要求进一步稀释。
保存：人β-神经生长因子,CHO表达(b NGF CHO)冻干粉末在室温可保存3周，或置于-18℃冻存用于长期保存。重新溶解后可以在4℃冷藏保存至少2-7天以上；如果想延长保存时间，建议分装后置于-18℃冻存。请避免反复冻融。

Introduction 

NGF-beta has nerve growth stimulating activity and the complex is involved in the regulation of growth and the differentiation of sympathetic and certain sensory neurons. Mutations in this gene have been associated with hereditary sensory and autonomic neuropathy, type 5 (HSAN5), and dysregulation of this gene's expression is associated with allergic rhinitis.

Description 

Nerve Growth Factor-beta Human Recombinant produced in CHO is a noncovalently disulfide linked homodimer, glycosylated, polypeptide chain containing 2 identical 118 amino acids and having a molecular mass of 26.5 kDa.
The NGF-b is purified by proprietary chromatographic techniques.

Physical Appearance 

Sterile Filtered White lyophilized (freeze-dried) powder.

Formulation 

The protein was lyophilized from a 0.2µm filtered solution in 20mM Tris-HCl pH-8 & 0.15M NaCl.

Solubility  （溶解性）

It is recommended to reconstitute the lyophilized NGF-b in sterile 18MΩ-cm H2O not less than 100µg/ml, which can then be further diluted to other aqueous solutions.

Stability    （稳定性）

Lyophilized Nerve Growth Factor b although stable at room temperature for 3 weeks, should be stored desiccated below -18C. Upon reconstitution Nerve Growth Factor-beta should be stored at 4C between 2-7 days and for future use below -18C.
For long-term storage it is recommended to add a carrier protein (0.1% HSA or BSA).
Please prevent freeze-thaw cycles.
