HCV NS3蛋白酶可用于在NS3/NS4A, NS4A/NS4B, NS4B/NS5A和NS5A/NS5B位点切割非结构蛋白。HCV NS3是一种胰凝乳蛋白酶样丝氨酸蛋白酶，需要结合辅因子－含54个氨基酸的NS4蛋白，以达到最佳活性。X射线晶体结构研究显示NS3可与NS4形成紧密的非共价结合物。NS3/4A蛋白酶是病毒复制和感染性病毒颗粒生成的关键蛋白酶，是抗-HCV药物治疗最关键筛选靶。重组HCV NS3/4A蛋白酶具有高活性，可与SensoLyte® 490, 520, 或 620 HCV蛋白酶检测试剂盒共同使用（Cat# 71126, 71145, 71146）去筛选anti-HCV蛋白药物。
HCV NS3/4A蛋白酶贮藏在20 mM 盐酸中，pH 8.0, 20% 甘油, 100 mM KCl, 1 mM DTT和0.2 mM EDTA.
