RoboSep™ Kit for Human CD4+CD127lowCD49d-CD25high Regulatory T Cells with Filter Tips is a simple two-step method that is designed to isolate CD4+CD127lowCD49d-CD25+ cells from whole blood or buffy coat. CD4+CD127lowCD49d- T cells are enriched from whole blood or buffy coat by negative selection using RosetteSep™ CD4+CD127lowCD49d- T Cell Pre-Enrichment Cocktail. The RosetteSep™ cocktail contains a combination of antibodies that is directed against non-CD4+CD127lowCD49d- T cells and glycophorin A on red blood cells (RBCs). When centrifuged over a buoyant density medium such as RosetteSep™ DM-L (Catalog #15705) or Ficoll-Paque™ PLUS (Catalog #07957), the unwanted cells pellet along with the RBCs. The purified cells are present as an enriched population at the interface between the plasma and the buoyant density medium. The enriched cells are then selected for CD25+ cells by positive selection using EasySep™ Human CD25 Positive Selection Kit. Cells are targeted with Tetrameric Antibody Complexes recognizing CD25 and dextran-coated magnetic particles. Labeled cells are separated using an EasySep™ magnet without the use of columns. Cells of interest remain in the tube while unwanted cells are poured off.

Product stable at 2 - 8°C until expiry date as indicated on label. Contents have been sterility tested. Do not use cocktail if vial contents have leaked. Unused cocktail may be disposed of according to standard laboratory procedures for non-hazardous liquids. Do not freeze this product. This product may be shipped at room temperature (15 – 25°C), and should berefrigerated upon receipt.
