激活素(activins) Activin A是转化生长因子(TGF)β家族的一种分泌蛋白，可控制胚胎轴发育为有功能的前肠源性组织，调解多种细胞的生长和分化。多种研究表明，其能诱导胰腺β细胞的分化。尤其与其他因子的联合作用后能发挥更显著的诱导作用。如activin A联合全反式维甲酸可以更有效地诱导鼠ESC为产胰岛素细胞，而activin与BTC共同作用可以转化新生大鼠导管细胞为产胰岛素细胞。卵泡抑素(follistatin)和表皮生长因子相关肽(Cripto)是ac2tivins的天然拮抗剂，有研究表明了胰岛分化阶段导管上皮细胞里的activins及其抑制因子follistatin和Cripto都有所增加,卵泡抑素抑制activin的同时，能显著促进胰腺导管细胞新生而减少β细胞的分化，故选择抑制activin信号途径可运用于扩增前体细胞，而激活activin有利于β细胞的分化。这些结果表说明,胰腺生长和分化之间的平衡可通过activin信号途径的精确调节来实现。同时它还被广泛用于干细胞研究，用来诱导分化的胚胎干细胞形成内胚层。

Activin A is a member of the TGFβ superfamily. It is a 26 kDa disulfidelinked homodimer of two βA chains, each containing 116 amino acid residues. Activin A is widely used in stem cell research. It has been used to induce the differentiation of embryonic stem cells to the definitive endoderm. The biological activities of Activin A can be neutralized by diffusible TGF-β antagonists, Follistatin (also available from StemRD). StemRD’s Activin A is made in human 293 cells, which renders human-specific post-translational modifications. We purified this product through a series of chromatography including HPLC to near homogeneity.

• Purity: Greater than 95% by SDS-PAGE and HPLC analysis
• Biological Activity: The activity was determined by using a SMAD reporter gene assay in human 293 cells. The EC50 ranges from 0.5 – 2.0 ng/ml
